Description

Organization Overview

We work on the cell cycle using budding yeast as a model system. We are collaborating to construct ordinary differential equation-based models for control of the yeast cell cycle, and to test these models genetically and biochemically. We are engaged in a combined biochemical and genetic screen for binding partners and phosphorylation targets specific to different cyclin-Cdk complexes. We plan a combined computational and molecular approach to analysis of promoter elements important in both the mitotic and meiotic cell cycles.
Overview

The overall purpose of the project is to search for broadly cross-reactive single-chain antibodies against viruses in the coronavirus family. Specifically the work will consist of assisting in: construction of libraries of yeast expressing single-chain antibodies against multiple viral antigens in the coronavirus family; screening the libraries for binding and cross-binding to antigens from related viruses; DNA preparation and analysis of next-generation sequencing data to determine sequences and specificities of cross-binding vs. specific antibodies.

Responsibilities

Responsibilities include:

  • Prepare plasmid and PCR-generated DNA for construction of yeast libraries

  • Carry out high-efficiency yeast transformation to generate complex libraries

  • Select differential binding clones using magnetic bead technology

  • Purify DNA from selected populations, prepare for next-generation sequencing

  • Aid with biochemical and structural characterization of resultant single-chain antibodies
  • Perform related duties and responsibilities as assigned/requested

Qualifications

Bachelor of Science degree with basic laboratory experience is required. Specific experience in molecular biology and microbiology (DNA preparation and analysis, media preparation, microbial culture) is strongly preferred.

The Rockefeller University is an equal opportunity employer – veterans/individuals with disabilities. Qualified applicants will receive consideration for employment without regard to characteristics protected by applicable local, state, or federal law, including but not limited to disability and protected veteran status.

The salary of the finalist selected for this role will be set based on various factors, including but not limited to organizational budgets, qualifications, experience, education, licenses, specialty, and training.
